is it possible to change the security permission (i.e. changing access to some drives) without restartting the PC?? coz it's quite time consuming like if a customer have a flash drive i want to allow him to access that i eventually reboot the PC for the changes to take effect..

No, because this permissions are based on windows registry and applied by system on startup.
One way to speed up process is logging off PC instead full reboot.
